Phase II Trial of Bortezomib and Vorinostat in Mantle Cell and Diffuse Large B-Cell Lymphomas

Study Description

Bortezomib and vorinostat may stop the growth of cancer cells by blocking some of the enzymes needed for cell growth. Giving bortezomib together with vorinostat may kill more cancer cells.

The purpose of this phase 2 trial is to study how well giving bortezomib together with vorinostat works in treating patients with recurrent mantle cell lymphoma or recurrent and/or refractory diffuse large B-cell lymphoma.

Patients will receive Vorinostat 400 mg (total daily dose as a single dose) on days 1-5 and days 8-12. Patients will receive Bortezomib 1.3 mg/m2/d IV on days 1, 4, 8, and 11.  The regimen is repeated every 3 weeks.

Patients who experience a complete or partial response after at least one year of treatment may go off  treatment while remaining on the study, and then resume study treatment upon disease progression if the study remains open.

Key Eligibility

  • Men and women age 18 and older
  • Diagnosed with 1 of the following:
    • Mantle Cell Lymphoma
      • Previously treated or untreated
    • Diffuse Large B-Cell Lymphoma
      • Have had at least 1 prior systemic therapy
      • No prior treatment with bortezomib
  • More detailed eligibility discussed when you contact the study team
